2020 USRA - Expression and isotope-labelling of membrane proteins for spectroscopic studies
Job Information
Expression and isotope-labelling of membrane proteins for spectroscopic studies
Proposed Start Date: 4 May 2020
Supervisor
Leonid Brown, Professor
Department: Physics
Brief Outline of Proposed Research Project
Our group studies energy and signal transduction as well as ion and water transport across biological membranes performed by membrane proteins using combination of molecular biology, biochemistry, and advanced spectroscopy. The incumbent will help in sample preparation for spectroscopic studies of membrane proteins.
Job description (task/responsibilities, relevant scheduling details, and required and/or preferred qualifications):
Protein expression, isotope and spin labelling (NO RADIOACTIVITY involved), lipid reconstitution. Techniques and instrumentation: growth of E. coli in shaker incubators, cell disruption by sonication, detergent solubilization, His-tag purification, SDS-PAGE, centrifugation, spectrophotometry, lipid reconstitution, ultracentrifugation, cysteine labeling, preparation of samples for mass spectrometry, FTIR, Raman, EPR, and NMR.
