How does a molecular switch work in hormone activated signalling?

Prevailing thinking describes GPCRs as loosely coupled "switches" implying that stochastic intramolecular processes somehow contribute to signalling and hormones somehow influence the connectivity or tightness of the switch. We describe a network of stabilizing interactions which may be reinforced by so-called inverse agonists, thereby keeping the switch "off" or weekend by the addition of partial and full agonists such that the protein can stochastically access activation intermediates and undergo signalling.
